Motigut 10 mg

Tablet

Generic: Domperidone

Square Pharmaceuticals PLC.

Therapeutic Class: Antiemetic & Prokinetic

Indications

Relief of nausea and vomiting. Also used for symptoms of delayed gastric emptying such as fullness after meals, bloating, belching and gastro-oesophageal reflux.

Pharmacology

Domperidone is a dopamine D2-receptor antagonist. It acts on the chemoreceptor trigger zone and on the upper gastro-intestinal tract, speeding gastric emptying and increasing the tone of the lower oesophageal sphincter. It crosses the blood-brain barrier poorly, so central side effects are uncommon.

Dosage & Administration

Adult and adolescent 12 years and over (35 kg or more): 10 mg up to 3 times daily, 15 to 30 minutes before meals; maximum 30 mg daily. Child under 12 years or under 35 kg: 0.25 mg/kg up to 3 times daily; maximum 0.75 mg/kg daily, as directed by the physician. Use the lowest effective dose for the shortest time, usually not more than 1 week.

Side Effects

Dry mouth, headache, abdominal cramps and diarrhoea. Rarely raised prolactin with breast enlargement or milk secretion, menstrual disturbance, abnormal movements and heart rhythm disturbance (QT prolongation).

Precautions & Warnings

Use with caution in kidney impairment, in the elderly and in patients with electrolyte disturbance. Avoid with medicines that prolong the QT interval or strongly inhibit CYP3A4, such as ketoconazole, erythromycin and clarithromycin. Use in pregnancy and breast-feeding only if advised by the physician.

Contraindications

Hypersensitivity to domperidone. Prolactin-releasing pituitary tumour (prolactinoma). Gastro-intestinal bleeding, obstruction or perforation. Moderate or severe liver impairment. Known QT prolongation, significant electrolyte disturbance or heart disease such as heart failure.

Storage Conditions

Store below 30°C in a cool, dry place, away from light. Keep out of the reach of children.
